چکیده

o-Dichlorobenzene (o-DCB) is used as an organic solvent, as a chemical intermediate, and as a heat transfer medium. In humans, o-DCB is metabolized to 2,3and 3,4-dichlorophenols, and 3,4and 4,5-dichlorocatechols, and these metabolites are eliminated via the kidneys. In this study, we tried to determine the concentrations of urinary 2,3and 3,4-dichlorophenols using a gas chromatograph (GC). When control urine specimens were spiked at concentrations of 10, 20 and 40 mg/l, the mean recovery rates of 2,3and 3,4-dichlorophenols were 98.3 to 101.9% (CV=4.0 to 4.8%) and 100.6 to 105.4% (CV=2.5 to 7.0%), respectively. Next, urine samples collected from ten male workers exposed to o-DCB were analyzed. The concentrations of urinary 2,3and 3,4-dichlorophenols determined by the GC method closely agreed with those by the HPLC method, which we had developed in a previous study, and these metabolite concentrations were linearly correlated to the 8h TWA values of o-DCB in the range of 0.1 to 2.3 ppm. Consequently, the GC method can be used for biological monitoring of o-DCB, though it is necessary that the linear relation is confirmed in a higher range of exposure.

Differences in urinary monochlorobenzene metabolites between rats and humans.

The high performance liquid chromatographic method for the determination of p-chlorobenzene mercapturic acid and 4-chlorocatechol conjugates is described. For determination of urinary mercapturic acid, the benzene extract from urine was injected into a liquid chromatograph and for determination of urinary 4-chlorocatechol conjugates, hydrolysate was dissolved in methanol. The methanol solution ...

Development and Validation of a GC-FID Method for Diagnosis of Methylmalonic Acidemia

Background: Urinary organic acids are water-soluble intermediates and end products of the metabolism of amino acids, carbohydrates, lipids, and a number of other metabolic processes. In the hereditary diseases known as organic acidurias, an enzyme or co-factor defect in a metabolic pathway leads to the accumulation and increased excretion of one or more of these acidic metabolites. Gas chromato...

Quantitative determination of urinary metabolites in subjects exposed to organic solvents.

Most of the common organic solvents are excreted into urine as metabolites. A correlation exists for several organic solvents between the amount taken in and the amount of metabolites excreted. Many methods have been developed for the measurement of these urinary metabolites. The methods were classified into three group: colorimetry, gas chromatography and high performance liquid chromatography...

Evaluation of a benchtop capillary gas chromatograph-mass spectrometer for clinical toxicology.

We evaluated the Hewlett-Packard 5995B benchtop capillary gas chromatograph-mass spectrometer (GC-MS) for its ability to identify drugs commonly detected and (or) measured in the clinical toxicology laboratory. Initial experiments indicated that the instrument as originally configured, with an isolation valve between the gas chromatograph and mass spectrometer, was unsatisfactory for the identi...

Multiresidue determination of organophosphorus pesticides in corn oil using solid-phase extraction with lanthanum silicate sorbent and gas chromatography

Phosalone, malathion and diazinon were analyzed in corn oil using solid phase extraction (SPE)with lanthanum silicate as a new solid sorbent followed by gas chromatography with nitrogenphosphorus detection (GC-NPD). The efficiency of different eluents (methanol, dichloromethane, ethyl acetate, dichloromethane — ethyl acetate (1:1) and ethyl acetate — methanol (1:1)) werecompared for elution of o...
